Zijn klanten gaan van voetbalmoeders tot topkoks, maar nooit kinderen -- daar stelt hij de grens. Wat zou er dus mis kunnen lopen? Veel. Hij geeft er uiteraard de voorkeur aan zich gedeisd te houden, maar leert dat geen goede daad ongestraft blijft als hij probeert wat plaatselijke tieners te helpen en besprongen wordt door drie dakloze punkers. Ze stelen zijn dope en zijn geld, wat hem in diepe schulden brengt bij zijn leverancier Brad (Ed Helms.) Om met een schone lei te herbeginnen -- en zijn goede gezondheid te behouden -- moet David een grote drugssmokkelaar worden door Brads laatste lading uit Mexico binnen te brengen. Mits een gezonde dosis overredingskracht weet David zijn buren voor zijn kar te spannen. De cynische stripper Rose (Jennifer Aniston), Kenny (Will Poulter), die altijd al meer praat had dan dat ie drugs kocht en Casey (Emma Roberts), een tiener vol tattoos en piercings, vormen in zijn waterdichte plan de "Millers". Eén nepvrouw, twee namaakkinderen en een blinkende motorhome later, trekken de Millers de grens over voor een verlengd weekend dat belooft te eindigen met het nodige spektakel. David Burke (Jason Sudeikis) est un dealer à la petite semaine qui se contente de vendre sa marchandise à des chefs cuisiniers et des mamans accompagnant leurs fils au football, mais pas à des ados -- car, au fond, il a quand même des principes ! Alors que tout devrait se passer au mieux pour lui, les ennuis s'accumulent... Préférant garder profil bas pour des raisons évidentes, David comprend, à son corps défendant, qu'on peut subir la pire injustice même lorsqu'on est animé des meilleures intentions : tentant de venir en aide à des jeunes du quartier, il se fait agresser par trois voyous qui lui volent sa marchandise et son argent. Il se retrouve dans une situation des plus délicates puisqu'il doit désormais rembourser son fournisseur, Brad (Ed Helms). Afin d'éponger sa dette -- et de rester en vie --, David n'a d'autre choix que de jouer dans la cour des grands en se rendant au Mexique pour ramener une importante cargaison de drogue à Brad. Réussissant à convaincre ses voisins -- Rose, une strip-teaseuse cynique (Jennifer Aniston), Kenny, qui aimerait bien tester la marchandise (Will Poulter) et Casey, une ado débrouillarde couverte de tatouages et de piercings (Emma Roberts) -- de lui venir en aide, il met au point un plan censé être infaillible : avec ses complices qu'il fait passer pour sa femme et ses deux grands enfants, il met le cap sur le Mexique au volant d'un camping-car flambant neuf le jour de la fête nationale. In 2005, insecure bookish student Morton Schmidt (Jonah Hill) and popular underachieving jock Greg Jenko (Channing Tatum) miss their school prom. Seven years later, the two meet again at Police Academy and become firm friends and partners on bicycle patrol. They catch a break when they arrest Domingo (DeRay Davis), the leader of a one-percenter motorcycle gang, but are forced to release him after Jenko failed to read him his Miranda rights. The two are reassigned to a revived scheme from the 1980s which specialises in infiltrating high schools. Captain Dickson (Ice Cube) gives them new identities and sends them to a high school to contain the spread of a synthetic drug called HFS. After mixing up their identities, Schmidt and Jenko are enrolled in classes ill-fitting to their personalities. Schmidt gets a lead on HFS from classmate Molly (Brie Larson) and he and Jenko meet the school's main dealer, popular student Eric (Dave Franco). The two take HFS in front of him to maintain their cover and after experiencing the drug's effects clash heads over how much school has changed since they left: Schmidt's intelligence makes him popular while Jenko's lax attitude is frowned upon. Eric takes a liking to Schmidt, who develops a romantic interest in Molly, while Jenko becomes friends with the students in his AP Chemistry class. Schmidt throws a party to gain Eric's trust. At the party, Jenko's friends hack Eric's phone and overhear information about an upcoming meeting between Eric and his supplier, but also catch Schmidt making disparaging comments about Jenko. The rift between the two grows as their new school life invades upon their official police work. Schmidt and Jenko track Eric to a money exchange with the distributors of HFS - the motorcycle gang from the park - and a chase ensues on the freeway. Schmidt hesitates firing his gun upon the gang members and he and Jenko return to school arguing, the ensuing fight interrupting the school play. The two are expelled from school and are removed from the Jump Street program. A stressed and terrified Eric later approaches Schmidt and Jenko retelling the freeway encounter, unaware of their role in it. He recruits them as security for a deal taking place at the school prom. At the prom, Schmidt finds Molly doped up on HFS: he reveals his true identity and begs her to leave. At the deal, Schmidt and Jenko are surprised find that the supplier is the physical education teacher Mr. Walters (Rob Riggle), who blackmailed Eric into being his dealer after finding him smoking marijuana. The motorcycle gang arrives for the deal but Molly interrupts, revealing that Schmidt is a cop. A Mexican standoff ensues in which Domingo orders his men to kill Schmidt and Jenko. Two of the gang members reveal themselves as undercover DEA agents Tom Hanson and Doug Penhall (Johnny Depp and Peter DeLuise reprising their original roles). In the ensuing gunfight Hanson and Penhall are fatally wounded. Mr. Walters and Eric escape in a limo with the money and Molly as a hostage; the gang, and Schmidt and Jenko, follow close behind. As the car chase winds through the city, Jenko creates a homemade bomb using knowledge garnered from chemistry classes and tosses it into the one-percenters' limo, blowing it up. Mr. Walters pulls over and attempts to flee on foot with Molly as his hostage. He shoots at Schmidt but Jenko takes the bullet in his arm, saving his partner's life; in response, Schmidt shoots Mr. Walters, unintentionally severing his penis. Jenko successfully reads Walters his Miranda Rights as he arrests both the teacher and Eric, and the two cops reconcile their relationship. A low-level marijuana drug dealer David Clark (Jason Sudeikis) living in Denver is robbed of his money and stash, some of which he owes to his wealthy drug lord supplier Brad Gurdlinger (Ed Helms). After David is brought to him, Gurdlinger forces David to smuggle marijuana from Mexico as a solution to clear his debt. Realizing that one man attempting to get through customs is too suspicious, he hires a stripper neighbor stage-named Rose (Jennifer Aniston), a runaway 21-year-old petty thief named Casey (Emma Roberts), and his 18-year-old neighbor Kenny (Will Poulter) to pose as a bogus family called the "Millers". When the Millers reach the compound, they find out that the amount of marijuana they were picking up is not "a smidge" as Gurdlinger had informed them, but actually two tons. Because of the extra load of the marijuana on the RV, one of the radiator hoses breaks while going up a steep incline. A family they had encountered at the border called the Fitzgeralds, consisting of Don (Nick Offerman), Edith (Kathryn Hahn), and Melissa (Molly Quinn), catch up to them and tow the Millers's RV to a repair shop. On the trip to the shop, David learns that Don Fitzgerald is a DEA agent after finding his badge and gun in the glove compartment.
The marijuana turns out to be stolen from Pablo Chacon's cartel where Gurdlinger had David pose as an agent of Chacon. The next day, when the Millers head to the shop to pick up the RV, Chacon (Tomer Sisley) and his henchman One-Eye (Matthew Willig) are waiting for them and prepare to execute the family. They immediately tell Chacon that they aren't a real family and that they didn't know they were stealing from him. Rose is given a chance to prove that she is a stripper by dancing, and when she gets close, turns a steam vent onto Chacon. The Millers then escape in the RV, with Kenny behind the wheel. Due to Kenny's erratic driving, the RV veers off the highway and a tarantula, hiding in a bowl of fruit given to them when they picked up the marijuana, crawls up Kenny's leg and bites his testicle.
As Kenny has a severe allergic reaction to the bite, the Millers head to the hospital. This further delays the delivery of the contraband, but David renegotiates with Gurdlinger for a fee of $500,000 on the condition that it arrives that night after mentioning on how he encountered Pablo Chacon. When Kenny is finally released, David rushes him to the RV in a wheelchair and accidentally tips him over. In the ensuing argument, David inadvertently reveals how much he is getting paid, in comparison to how little he offered to pay each of the others. Casey, Rose, and Kenny are left in disgust by the revelation, and so David leaves them at the local carnival.
Shortly after leaving, David regrets abandoning them after hearing "Waterfalls" by TLC on the radio and returns to the carnival, begging them to come back with him. On their way to the RV, they encounter Edith and Melissa. One-Eye discovers them and is about to shoot everyone, but Don comes out of his camper and subdues him. Chacon then comes around the corner with Melissa held at gunpoint and is about to kill them all, but is momentarily distracted by the 4th of July fireworks. David and Kenny subdue him. Don arrests Chacon and One-Eye, but lets the Millers leave.
David delivers the drugs to Gurdlinger, who tells him he's late and that their deal is off, but then admits that he had never intended to pay at all. DEA agents crash into the room, arresting Gurdlinger and his henchmen. The agent in charge is Don who tells David that he will have to be in the witness protection program until the trial of Gurdlinger and Chacon. He then adds that anyone that was a witness to the crime will be in protection and David smiles. The Millers are then seen together in a suburbian house, with several marijuana plants growing in the garden.
